You can park your car on the Canadian side, walk into the US side and proceed to US customs, tell them you're there for a NEXUS interview. They will clear you through the usual clearance process asking the typical questions, i.e. what are you doing here, how long will you be here, any goods, weapons/firearms to declare etc. Having said that, it's always better to be safe than sorry so I would personally suggest you drive across. (More on that shortly.)
Once clear, make your way to the NEXUS office. The officers there will tell you where to go. From that point, it's pretty smooth sailing. The NEXUS office is pretty quiet for the most part so you may or may not have to wait very long for the interview, hence my suggestion to arrive early. The interview process itself is pretty quick and easy anyway. So let's say your interview is scheduled for 2pm, you arrive at 1:15pm to be safe, if they aren't busy (and most likely they won't be) they will slip you in rather than have you waiting. After all, they don't want people in their face so they want you in and out as quick as possible. Not to mention, they would rather have candidates show up early than be late. Being late should be your biggest fear.
I did my renewal there once and also picked up a set of Travel Declaration Cards (TDC) on a seperate occassion. I was in and out in less than 15 minutes. Mind you, this was over 5 years ago.
